Built between 1526 and 1542. The walls were made of adobe bricks and plastered with mud (talpuja), and the roof was covered with clay tiles. The current area is 748 m².

In 1526, Mercedarian monks built it as a simple hut. It was considered the first building in service to the Spanish Crown. In 1532, it was rebuilt with adobe brick walls and a tile roof. Two to three masses were held on weekends, accompanied by musical instruments.

During archaeological investigations in 2000, a crypt was found near the main altar in the church, possibly containing the remains of Captain Francisco Hernández de Córdoba and Governor Pedrarias Dávila.
